Principle of the Assay: Human EDIL3 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for EDIL3 has been precoated onto 96-well plates. Standards(Expression system for standard: NSO; Immunogen sequence: D24-E480) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for EDIL3 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin- Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human EDIL3 amount of sample captured in plate.

Background/Introduction: EDIL3, also known as DEL1, is a glycoprotein that is secreted by endothelial cells and can associate with the endothelial cell surface and the extracellular matrix. It is regulated by hypoxia or vascular injury and has been implicated in vascular remodeling during angiogenesis. The EDIL3 gene is mapped to 5q14. Human endothelial cells bind to Del1 together with the alpha-V (ITGAV)/beta-3 (ITGB3) integrin receptor. It is confirmed that adhesion of endothelial cells to Del1 is integrin mediated and that Del1 binds ITGAV/ITGB3. Functional analysis showed that both Del1 and vitronectin (VTN) stimulate endothelial cell migration. Immunoprecipitation and immunoblot analyses revealed that adhesion of endothelial cells to Del1 induces tyrosine phosphorylation of PTK2
